چکیده

Using crude extracts from the cyanobacterium Anabaena variabilis glutamine synthetase (GS) activity was rapidly irreversibly reduced to about 60% during dark incubation („sponta neous GS inactivation“). An additional decrease was observed by the addition of ammonia in the light („ammonia-mediated inactivation“). Both effects were prevented by EDTA, MnCl2 or catalase indicative of the involvement of H20 2. This is a key intermediate in oxida tive enzyme inactivation. In both spontaneous and ammonia-mediated GS inactivation H20 2 is produced in different ways. Spontaneous inactivation is prevented by depletion of reduced pyridine nucleotides which apparently donate electrons to produce H 20 2. Fractionation of the crude extract showed that the light-enhanced GS inactivation by ammonia required the presence of thylakoid membranes. The photosynthesis inhibitor DCMU decreased GS inacti vation by ammonia. For the inactivation in the light apparently H20 2 is produced from super oxide during photosynthetic electron transport.

Oxidative modification of glutamine synthetase by amyloid beta peptide.

beta-Amyloid peptide (A beta), the main constituent of senile plaques and diffuse amyloid deposits in Alzheimer's diseased brain, was shown to initiate the development of oxidative stress in neuronal cell cultures. Toxic lots of A beta form free radical species in aqueous solution. It was proposed that A beta-derived free radicals can directly damage cell proteins via oxidative modification. Re...

Oxidative Modification of Glutamine Synthetase

The first step in the proteolytic degradation of bacterial glutamine synthetase is a mixed function oxidation of one of the 16 histidine residues in the glutamine synthetase subunit (Levine, R. L. (1983) J. Biol. Chem. 258, 11823-11827). A model system, consisting of oxygen, a metal ion, and ascorbic acid, mimics the bacterial system in mediating the oxidative modification of glutamine syntheta...

Formation of Hydrogen Peroxide by Chilled Goat Spermatozoa and the Effects of Dead Spermatozoa on Motility Characteristics

In this study, the formation of hydrogen peroxide (H2O2) by chilled goat spermatozoa was measured. Furthermore, the effects of dead spermatozoa on motility characteristics were studied. Fresh collected ejaculates from five Shami bucks were centrifuged and virtually all seminal plasma was removed. A part of the collected spermatozoa was killed by two ways: the first by repe...

The effect of eight weeks of aerobic exercise and coriander seed extract on oxidative stress and cellular energy indices of heart tissue in male rats poisoned with hydrogen peroxide

Introduction: The aim of this research was to investigate the effect of eight weeks of aerobic training and coriander seed extract with two doses of 5 and 10 mg on oxidative stress markers and cardiac muscle ATP levels of rats poisoned with hydrogen peroxide. Methods: 42 healthy adult male rats were randomly divided into 7 groups. During one week, the groups were poisoned intraperitoneally by ...

Inhibition by dithiothreitol of the utilization of glutamine by carbamyl phosphate synthetase. Evidence for formation of hydrogen peroxide.

The glutaminase activity of glutamine-dependent carbamyl phosphate synthetase (Escherichia coli) and that of the separated light subunit of this enzyme, as well as the glutaminedependent synthetase activity catalyzed by the native enzyme, are inactivated by incubation in air with relatively low concentrations (0.025 to 1 mM) of dithiothreitol; oxidized dithiothreitol does not inhibit. Other mer...
